A folyamatban fontos szerepet j\u00e1tsz\u00f3 sonic hedgehog (<strong>Shh)<\/strong> molekula a fejl\u0151d\u0151 v\u00e9gtagkezdem\u00e9ny egyik speci\u00e1lis, polariz\u00e1ci\u00f3s z\u00f3na (<b>ZPA<\/b>&#8211; zone of polarizing activity) nev\u0171 r\u00e9gi\u00f3j\u00e1ban expressz\u00e1l\u00f3dik. A ZPA mezenhim\u00e1lis sejtek kis csoportja a v\u00e9gtagbimb\u00f3 poszterior r\u00e9sz\u00e9n, mely meghat\u00e1rozza a k\u00e9s\u0151bb keletkez\u0151 ujjak sz\u00e1m\u00e1t \u00e9s milyens\u00e9g\u00e9t. Hat\u00e1s\u00e1t hossz\u00fa t\u00e1von fejti ki, vagyis az apik\u00e1lis ektoderm\u00e1lis red\u0151 (<strong>AER<\/strong>) sejtjei, melyek reag\u00e1lnak a Shh-szign\u00e1lra, nem k\u00f6zvetlen\u00fcl a ZPA sejtek mellett helyezkednek el.<\/p>\n<p style=\"text-align: justify;\">A ZPA-val \u00e9s a Shh-gal kapcsolatban a hossz\u00fa t\u00e1v m\u00e1s szempontb\u00f3l is fontos keres\u0151sz\u00f3. Ugyanis a <b>ZRS enhanszer<\/b>, az a regul\u00e1l\u00f3 elem, ami meghat\u00e1rozza, hogy a Shh a ZPA sejtjeiben termel\u0151dj\u00f6n, mag\u00e1t\u00f3l a <i>shh<\/i> g\u00e9nt\u0151l t\u00f6bb, mint 1Mb (1 milli\u00f3 b\u00e1zisp\u00e1rnyi) t\u00e1vols\u00e1gra van, m\u00e9ghozz\u00e1 egy m\u00e1sik g\u00e9n (<i>lmbr1<\/i>) intronj\u00e1ban [1]. R\u00e1ad\u00e1sul a <i>shh<\/i> g\u00e9n \u00e9s a ZRS enhanszer k\u00f6z\u00f6tt t\u00f6bb funkcion\u00e1lis \u00e9s az egyedfejl\u0151d\u00e9s adott szakasz\u00e1ban nem akt\u00edv g\u00e9n tal\u00e1lhat\u00f3. \u00c9rdekes m\u00f3don a g\u00e9nek sorrendje \u00e9s az enhanszer elhelyezked\u00e9se szint\u00e9ni\u00e1t mutat az ember, az eg\u00e9r \u00e9s a fugu (<i>Fugu rubripes<\/i>) genomj\u00e1ban is. S\u0151t, mi t\u00f6bb, az eg\u00e9rben azonos\u00edtott cisz-regul\u00e1l\u00f3 elemmel homol\u00f3g hal enhanszer transzg\u00e9n eg\u00e9r embri\u00f3kban k\u00e9pes volt rekapitul\u00e1lni az eredeti <i>shh<\/i> expresszi\u00f3t a fejl\u0151d\u0151 v\u00e9gtagban [1]. Hogy pontosan hogyan fejti ki specifikus hat\u00e1s\u00e1t az enhanszer ekkora t\u00e1vols\u00e1gon, arra van p\u00e1r elm\u00e9let \u00e9s persze p\u00e1r k\u00eds\u00e9rletes v\u00e1lasz, \u00e1m a hossz\u00fa t\u00e1v\u00fa cisz-regul\u00e1ci\u00f3 mechanizmusa pontosan m\u00e9g mindig nem tiszt\u00e1zott.<\/p>\n<p style=\"text-align: justify;\">Visszat\u00e9rve a jel\u00e1tvitelre, a v\u00e9gtagfejl\u0151d\u00e9sben is szerepet j\u00e1tsz\u00f3 szignaling molekul\u00e1k egy r\u00e9sze egy adott sejtcsoportban termel\u0151dik, hat\u00e1sukat viszont ezekt\u0151l viszonylag t\u00e1vol tal\u00e1lhat\u00f3 sejtekben fejtik ki. A jel\u00e1tviv\u0151k terjed\u00e9s\u00e9nek mechanizmus\u00e1ra sz\u00e1mos elm\u00e9let l\u00e9tezik, gerincesekn\u00e9l f\u0151k\u00e9nt a passz\u00edv diff\u00fazi\u00f3 \u00e9s a transzcit\u00f3zisnak nevezett akt\u00edv transzport az elfogadott \u2013 ez ut\u00f3bbi esetben a molekul\u00e1kat az arra nem v\u00e1laszol\u00f3 szomsz\u00e9dos sejtek felveszik, majd tov\u00e1bbadj\u00e1k. Gerinctelen embri\u00f3k eset\u00e9ben le\u00edrtak olyan mechanizmust, melyben speci\u00e1lis sejtny\u00falv\u00e1nyok (citon\u00e9m\u00e1k) sz\u00e1ll\u00edtj\u00e1k az adott jel\u00e1tviv\u0151t a felhaszn\u00e1l\u00f3 sejtekig. A Nature online kiadv\u00e1ny\u00e1ban vas\u00e1rnap megjelent publik\u00e1ci\u00f3 szerint a gerinces embri\u00f3kban a Shh is hasonl\u00f3 sejtny\u00falv\u00e1nyokon kereszt\u00fcl jut el a c\u00e9lsejtekig [2].<\/p>\n<p><!--more--><\/p>\n<p style=\"text-align: justify;\"><img loading=\"lazy\" decoding=\"async\" class=\"imgleft\" style=\"margin-right: 5px;\" src=\"http:\/\/m.cdn.blog.hu\/cr\/criticalbiomass\/image\/post_img\/2013\/04\/fig1.jpg\" alt=\"fig1.jpg\" height=\"277\" width=\"275\" \/>Sanders \u00e9s koll\u00e9g\u00e1i transzg\u00e9n csirkeembri\u00f3k v\u00e9gtagbimb\u00f3it vizsg\u00e1lt\u00e1k egy \u00fajfajta mikroszk\u00f3pos technika seg\u00edts\u00e9g\u00e9vel, mely lehet\u0151v\u00e9 tette sz\u00e1mukra, hogy az \u00e9l\u0151 embri\u00f3kr\u00f3l sejtszint\u0171, s\u0151t sejtalkot\u00f3szint\u0171 felbont\u00e1sban time lapse felv\u00e9teleket tudjanak k\u00e9sz\u00edteni. Az embri\u00f3k olyan fluoreszcens transzg\u00e9n konstrukci\u00f3t tartalmaztak, mely a fejl\u0151d\u0151 v\u00e9gtagbimb\u00f3 mezenhim\u00e1lis sejtjeinek membr\u00e1nj\u00e1t jel\u00f6lte meg. A mikroszk\u00f3pos k\u00e9pek egy, a mezenhim\u00e1lis sejtek v\u00e9kony ny\u00falv\u00e1nyaib\u00f3l \u00e1ll\u00f3, \u00f6sszef\u00fcgg\u0151 h\u00e1l\u00f3zatot mutattak. A funkcion\u00e1lis vizsg\u00e1latok alapj\u00e1n kider\u00fclt, hogy a v\u00e9gtagkezdem\u00e9ny filop\u00f3diak\u00e9pz\u0151 sejtjei k\u00f6z\u00f6tt vannak a Shh-termel\u0151 ZPA-sejtek is, melyek ny\u00falv\u00e1nyai el\u00e9rik az apik\u00e1lis ektoderm\u00e1lis red\u0151 (<b>AER<\/b>) Shh- illetve FGF-jelre v\u00e1laszol\u00f3 sejtjeit, \u00e9s az AER sejtjei is, melyek ny\u00falv\u00e1nyai a ZPA-sejtek fel\u00e9 ir\u00e1nyulnak.
