{"id":16324600,"date":"2021-03-18T10:42:09","date_gmt":"2021-03-18T10:42:09","guid":{"rendered":"http:\/\/criticalbiomass.hu\/?p=16324600"},"modified":"2021-03-18T10:42:10","modified_gmt":"2021-03-18T10:42:10","slug":"hullo-konnyek-organoid-zapora","status":"publish","type":"post","link":"https:\/\/criticalbiomass.hu\/?p=16324600","title":{"rendered":"Hull\u00f3 k\u00f6nnyek (organoid) z\u00e1pora"},"content":{"rendered":"\n<p class=\"wp-block-paragraph\">Az organoidok, vagyis miniat\u0171r, <em>in vitro <\/em>(l\u00e1sd m\u00e9g \u201cPetri cs\u00e9sz\u00e9ben&#8221;) l\u00e9trehozott sz\u00f6vetdarabok kutat\u00e1sa az ut\u00f3bbi \u00e9vekben mindenk\u00e9ppen a leggyorsabban fejl\u0151d\u0151 biol\u00f3giai tudom\u00e1nyter\u00fcletet jelenti \u00e9s manaps\u00e1g m\u00e1r t\u00e9nyleg (hogy elpuffantsuk az idek\u00edv\u00e1nkoz\u00f3 nagy k\u00f6zhelyet) nem m\u00falik el h\u00e9t 1-2 fajs\u00falyosabb organoidos publik\u00e1ci\u00f3 n\u00e9lk\u00fcl. <\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Mad\u00e1rt\u00e1vlatb\u00f3l n\u00e9zve, k\u00e9t nagy csoport k\u00fcl\u00f6n\u00edthet\u0151 el organoidok eset\u00e9ben: azok, amelyek nagyon sz\u00e9les differenci\u00e1ci\u00f3s potenci\u00e1llal rendelkez\u0151 \u201cpluripotens&#8221; (ak\u00e1r klasszikus, embrion\u00e1lis, ak\u00e1r \u00fajgener\u00e1ci\u00f3s induk\u00e1lt) \u0151ssejtekb\u0151l j\u00f6nnek l\u00e9tre \u00e9s azok, amelyek sz\u0171kebb potenci\u00e1llal rendelkez\u0151, \u201cmultipotens\u201d \u0151ssejtekb\u0151l alakulnak ki. Ut\u00f3bbiak d\u00f6nt\u0151en a feln\u0151tt, kifejlett egyedek egyes sz\u00f6veteiben jelen lev\u0151 \u0151ssejtek felhaszn\u00e1l\u00e1s\u00e1val l\u00e9trehozott organoidok \u00e9s ennek a ter\u00fcletnek az egyik legfontosabb \u00fatt\u00f6r\u0151je a p\u00e1r \u00e9ve <a href=\"https:\/\/breakthroughprize.org\/Laureates\/2\/L29\">Breakthrough Prize-al is kit\u00fcntetett<\/a> Hans Celvers.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><a href=\"https:\/\/www.hubrecht.eu\/research-groups\/clevers-group\/\">Clevers csoportja<\/a> a holland Hubrecht Institute-ban m\u00e9g a 2010-es \u00e9vek elej\u00e9n \u00e9rt el komoly \u00e1tt\u00f6r\u00e9seket a b\u00e9l-organoidok l\u00e9trehoz\u00e1s\u00e1val \u00e9s kutat\u00e1s\u00e1val, az\u00f3ta pedig, tal\u00e1n csak minim\u00e1lis t\u00falz\u00e1ssal, nincs olyan <a href=\"https:\/\/hu.wikipedia.org\/wiki\/H\u00e1msz\u00f6vet\">h\u00e1msz\u00f6vettel (epit\u00e9liummal)<\/a> bor\u00edtott szerv, amelyb\u0151l ne k\u00e9sz\u00edtettek volna organoidokat.  Az ut\u00f3bbi \u00e9vekben k\u00fcl\u00f6nb\u00f6z\u0151 mirigyekre kezdtek koncentr\u00e1lni, ehhez kapcsol\u00f3dott egy tavalyi publik\u00e1ci\u00f3juk, ahol p\u00e9ld\u00e1ul <a href=\"https:\/\/www.sciencedirect.com\/science\/article\/pii\/S0092867419313236\">m\u00e9reg-termel\u00e9sre k\u00e9pes k\u00edgy\u00f3 m\u00e9regmirigy-organoidokat<\/a> csin\u00e1ltak. Ha a sejtek k\u00f6z\u00f6tt vannak \u0151ssejtek is, akkor a megfelel\u0151 k\u00f6zeg kik\u00eds\u00e9rletez\u00e9se ut\u00e1n el\u0151bb-ut\u00f3bb megjelennek a labda alak\u00fa organoidok. Most is ez t\u00f6rt\u00e9nt, el\u0151bb emberi, majd eg\u00e9r k\u00f6nnymirigyb\u0151l kiindulva hozt\u00e1k l\u00e9tre ezeket az organoidokat (al\u00e1bb csak az eg\u00e9r esetet mutatom), amelyek kb. 10 nap alatt \u223c200\u00a0\u03bcm \u00e1tm\u00e9r\u0151j\u0171 g\u00f6mb\u00f6kk\u00e9 n\u0151tt\u00e9k ki magukat \u00e9s a sz\u00f6vetteny\u00e9szetek fenntart\u00e1s\u00e1ra klasszikusan haszn\u00e1lt \u201cpassz\u00e1l\u00e1sok\u201d (\u00fajabb teny\u00e9szet ind\u00edt\u00e1sa a sejtekb\u0151l) ut\u00e1n is megmaradt ez a jelleg (az \u00e1bra B panelj\u00e9n a \u201cp<em>X<\/em>\u201d azt jelenti, hogy <em>X<\/em> db. passz\u00e1l\u00e1s ut\u00e1n; a \u201c0\u201d azt jelenti, hogy a frissen l\u00e9trehozott sz\u00f6vetet jellemzik.) Norm\u00e1lis k\u00f6nnymirigyn\u00e9l a k\u00f6nny az elvezet\u0151dik, de az organoid egy z\u00e1rt rendszer, \u00edgy a megtermel\u0151d\u0151, az organoid belsej\u00e9be \u00fcr\u00fcl\u0151 k\u00f6nny lufik\u00e9nt \u201cfelf\u00fajja\u201d az organoidokat. Ezt a m\u00e9retv\u00e1ltoz\u00e1st pedig el\u00e9g k\u00f6nnyen lehet automatiz\u00e1lva is detekt\u00e1lni, s ahogy az a fenti \u00e1br\u00e1n is l\u00e1that\u00f3, el\u00e9g gyorsan lehet sz\u0171rni azokat az anyagokat, amelyek k\u00f6nnyez\u00e9st tudnak induk\u00e1lni.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Mi\u00e9rt lehet ez hasonl\u00f3, mi\u00e9rt akarn\u00e1k el\u00e9rni, hogy valaki k\u00f6nnyezzen (\u00e9s mi\u00e9rt nem h\u00e1moztatunk vele hagym\u00e1t)? A helyzet az, hogy b\u00e1r egy k\u00f6nnymirigy anat\u00f3miailag el\u00e9g egyszer\u0171 rendszer, m\u00e9g nem \u00e9rtj\u00fck teljesen, mik\u00e9pp m\u0171k\u00f6dik.
